I am M.Tech in Biotechnology completed in 2006 as fresher. I did my project on “Purification and Dose Response Studies of Recombinant Proteins (Transglutaminase, ALT-2 & Combination) of Brugia malayi as vaccine in experimental animals Against Human lymphatic Filariasis.”
Brief of project is as below.
An attempt made in my project was, to study the Dose response of recombinant Brugia malayi Transglutaminase (rBmTGA), as a suitable candidate vaccine against human lymphatic filariasis. The transglutaminase gene from Brugia malayi adult female parasite in pRSETB vector (T7 expression system) was expressed as 6-histidine tagged fusion protein and purified by Immobilized Metal Affinity Chromatography (IMAC).
The imunoreactivity of the protein was shown by Western Blot with Anti-histidine and Anti TGA antibody.

Thus purified recombinant protein TGA was immunized in mice, individually (TGA only) as well as in combination with another established filarial vaccine candidate ALT-2. We have optimized the dose of the antigen (TGA), which gives maximum antibody titre by ELISA method. Further we have also shown that immunization of TGA along with ALT protein gives higher titre of antiserum than immunization with TGA alone.
